PAGE 毕 业 论 文 ( 2012 届) 题 目 煤基活性炭固定化酶催化制备光学纯手性仲醇 煤基活性炭固定化酶催化制备光学纯手性仲醇 摘要:活性炭微孔发达、比表面积高、吸附能力强,是一种优良的吸附材料。“多孔”是活性炭的主要特征,正是由于多孔从而决定了活性炭的巨大的比表面积和超强的吸附性能。煤基大孔活性碳的表面官能团数目多,作为固定化酶载体的效果更佳。脂肪酶(EC) 由于其优良的化学、几何、立体选择性以及温和的反应条件,已经广泛应用于酶法拆分外消旋化合物以获得光学纯的手性物质。但应用游离酶酶不易回收重复使用及产物很难分离纯化,而采用酶固定化技术则可以克服以上不足。本文利用廉价的煤基活性炭吸附固定假单胞菌脂肪酶(Pseudomonas sp Lipase,PSL),获得固定化酶(PSL/AC)。微孔活性炭固定化脂肪酶酶在正庚烷溶剂中催化(R,S)-1-苯乙醇转酯化拆分反应,(S)-1-苯乙醇对映体过量值eeS和(R)-乙酸-1-苯乙酯的对映体过量值eeP均为98%,(R,S)-1-苯乙醇的转化率达到理论值。 关键词:活性炭,脂肪酶,手性仲醇,拆分 Coal-based functional enhancement and immobilized enzyme catalyzed the synthesis of optically pure Chiral secondary alcohols Abstract: Developed activated carbon micropores, High specific surface area, Adsorption ability, Is an excellent absorbent material, Porous is the main characteristics of activated carbon, It is due to the porous so as to determine the activated carbon huge surface area and strong adsorption properties.Has good compatibility to carbon and microbes, macroporous, coal-based activated carbon large surface area, narrow pore size distribution and adsorption capacity, adsorption rate was fast, acid, alkali, high temperature, etc. Widely used in the field of environmental protection, chemical, Macroporous activated carbon surface functional groups in coal-based number, the better effect of Immobilized Enzymes.Lipase (EC) due to its excellent chemical, geometric, stereoselective and mild reaction conditions, has been widely used in enzymatic resolution of racemic compounds in order to obtain optically pure chiral substances.Compared with chemical resolution law Enzymatic Resolution of a green and economical way.Application of the free enzyme is not easy recycling reuse and product are difficult to isolate purified using enzyme immobilization technology can overcome the above shortcomings.The enzyme immobilization technology in the most critical step is the preparation of performance, be able to match the carrier with the enzyme to achieve the high selectivity of the immob
