Agricultural College, Guizhou University, Guiyang Guizhou 550025, China) Abstract: Three methods w ere applied to extract total RN A from the leaves in order to understand the effects of leaf ages and genotypes and choose the best method for total RNA extraction in hot pepper in the present study. T he results show ed that the 28 S and 18 S bands extracted by method 2 w ere strong, and the uality and concentration of the total RNA were high. The lightness and uality of the total RNA from young leaves extracted by method 2 w ere the highest, next came medium leaves, and then old ones. T he 28 S and 18 S bands from the leaves of line 0860- 1 extracted by method 2 w ere bright, and total RNA concentration w as the highest; but those of the lines 0876 and 08122 w ere dark, and the concentration w as low .
