三角褐指藻生物学特性比较研究及转基因初步探索 Comparative study of characteristics of Phaeodactylum tricornutum and the preliminary exploration of genetically modified system Abstract Phaeodactylum tricornutum is a marine eukaryotic unicellular algae , a kind of very important primary producers of the marine ecological system. In addition, Phaeodactylum tricornutum with a high content of fatty acid, is a potential raw material for biodiesel.Therefore, Phaeodactylum tricornutum is often used as a scientific research material. Phaeodactylum tricornutum have different sources and forms, different ecological environments produce different effects. Phaeodactylum tricornutum are rich in unsaturated fatty acids with a variety of value, by means of genetic engineering ,they can be further improved in their content. This paper used Phaeodactylum tricornutum as the research object, compared the different strains of algae, and we had a preliminary exploration of one strain o f the algae in transgene. This experiment consists of two parts. The first part is the comparative study of 10 strains of Phaeodactylum tricornutum with different sources. Based on morphological and molecular identifications of the selected 10 strains, the 10 strains of algae were observed under a microscope, the morphological structure was observed.They were appeared the spindle, ovoid, and few were three radiate. The color of them was yellow-brown, they were morphological preliminarily identified as Phaeodactylum tricornutum. Genomic DNA of the 10 strains of algae were extracted and rbcL gene was amplificated, After sequencing the rb
