Abstract: As a new technique fo DNA finge p int ing, denatu ing g adient gel elect opho esis ( DGGE) can be used fo mic obial
dive sity analysis in envi onmental samples. In this study, PCRDGGE w as applied to analyze the mic obial communities in lake
sediments. Sediment samples f om seven locations in th ee lakes we e collected and thei genomic DNA w as di ectly ext acted. The
DNA yield of sediments of Lake Xuanwu and Lake Mochou was high ( 10gg) , while that of sediments in Lake Taihu was
elatively low . Afte DNA pu ification, the 16S RNA genes ( V3 to V5 egion) we e amplified with two unive sal p ime s
( F341GC and R907) . These amplified DNA f agments w e e sepa ated by pa allel DGGE.
The DGGE p ofiles indicate diffe ent bands patte ns fo diffe ent samples. The e a e five common bands in all lake sediment
samples, which indicate that the e a e simila ities among the populations of mic oo ganisms that exist in all lake sedim
