摘 要 野黄芩苷是黄芩茎叶中最为主要的化学成分,它具有降低脑血管阻力,改善脑血循环、增加脑血流量及抗血小板凝集的作用,已被制成针剂、药片、胶囊等多种剂型。目前此药物急缺,国内外市场已经不能满足,因此高纯度的野黄芩苷有待大规模的开发,野黄芩苷具有很高的利用价值,野黄芩苷产业具有良好的市场开发前景75%,提取温度:70℃,料液比:1:12,提取时间:3 h,提取次数:3次。在此条件下,黄芩茎叶总黄酮得率平均为1.39%,浸膏中总黄酮含量为21.02%。 第二,采用石油醚对提取液进行热回流脱脂实验,运用单因素试验考察碱提pH、酸沉pH、料液比等提取条件对黄芩茎叶总黄酮含量和浸膏得率的影响。采用合适的提取工艺得到野黄芩苷。碱提pH为7.5,酸沉pH为0.5,料液比为1:12,所得沉淀中总黄酮含量为80%以上。同时,以甲醇为溶剂进行重结晶从总黄酮中分离得到野黄芩苷,采用的方法操作简单,得到的野黄芩苷纯度较高,适合于工业化生产。 第三,建设年产20吨,90%野黄芩苷制备的初步的工业设计,并绘制带控制点的工艺流程图。 关键词:黄芩茎叶总黄酮;野黄芩苷;提取;纯化;工艺设计 Abstract Wild baicalin in scutellaria baicalensis stem leaf is the most main chemical composition, it can reduce cerebral vascular resistance, improve cerebral blood circulation, increase cerebral blood flow and antiplatelet aggregation effect, has been made into injection, tablets, capsules, etc. A variety of dosage forms. Based on scutellaria baicalensis stem leaf as raw material, extraction and purification of radix scutellariae stem Ye Ye baicalin were studied industrial design, and the capacity of 20 tons, 90% of wild baicalin production process has carried on the preliminary industrial design. First, to explore experiment, and screened wild baicalin extraction and the optimum process and technological parameters, the extraction technology parameters as follows: ethanol volume fraction: 75%, the extraction temperature: 70 ℃, material liquid than: 1:12, extracting time, 3 h, extraction times, 3 times.
