堆肥湯製作及菌群多樣性探討 Compost Tea Making and Bacterial Diversity during Fermentation 研究生:溫淑芬 Sue-Fen Wen 指導教授:邱瑞宇 Juei-Yu Chiu 【摘要】 現代化農業大量的使用農藥與化學肥料,導致土壤微生物相受到破壞,產生土壤酸化與污染的問題。為了永續農業發展,近年來堆肥湯 compost tea ,本實驗、、、以1噸醱酵探討兩種通氣方式(沉水馬達、魯式幫浦)及兩種菌群(市售混合菌、堆肥萃取液)對醱酵的影響由沉水馬達而有機質降解能力以堆肥萃取液降解率2.1 %及C/N共降低17.35皆高於市售,市售混合菌水溶性氮濃度增加量518 mg/L水溶性磷增加37 mg/L皆高於堆肥萃取液,由醱酵過程菌數顯示堆肥萃取液醱酵14天增加至109/mL高於市售混合菌,而堆肥萃取液真菌數醱酵21天為108 /mL高於市售混合菌生質量兩菌群皆至第13天為最高1.04-1.05。腐化參數以堆肥萃取液黃酸有機碳共增加80.3 %高於市售共增加43%,且以相關係數發現黃酸有機碳與空心菜有很的相關,因此推薦黃酸有機碳為產品品質參數。水耕空心菜栽培結果以,。為了解堆肥湯進行小型5L醱酵槽通氣醱酵,結果顯示堆肥萃取液蛋白質及基酸增加66 mg/L51 mg/L皆高於市售混合菌變性梯度膠體電泳法(enaturant gradient gel electrophoresis, DGGE)結果顯示市售混合菌經過醱酵10天後菌群只剩溶磷菌而堆肥醱酵至第4天菌群多樣性最高,顯示好氣醱酵使4天即可達到較高菌群多樣性 關鍵字:堆肥湯、變性梯度膠體電泳法、黃酸、基酸、多樣性 Modern agriculture utilizes large amount of chemical fertilizer and Pesticids. It destroys the microorganism diversity in the soil and causes acidification and pollution of the soil. For the sustanible agricultural, utilization of compost tea becomes popular but little research on it. In this experiment, the agricultural waste molasses added skim milk power、 milt and chitosan as compost tea making substance in 1ton’s fermentor. We look into the influence of two different ways of ventilation submersible motor , tubing pump and two kinds of Microbial consortium from commercial product, compost extract during fermentation. According to the analysis, submersible motor is better than tubing pump in terms of improving the value of DO, ORP, pH , EC, and the rise of temperature. Regarding the ability of biodegradation, the biodegradation rate of the compost extract inoculum is 2.1% and the C/N is 17%, both of them are higher than the commercial product inoculum. However, for the fertility efficiency, the commercial product inoculum compost tea increases 518mg/L of the concentration of nitrogen and 37mg/L of phosphorous, for which both are higher than inoculum compost extraction.
