矮型鸡于正常型鸡的miRNA表达谱分析

摘 要 在现代社会,肥胖问题已经成为全球性的一个公共健康疾病,引起了人们越来越多的关注。脂肪的生长涉及多基因表达、多信号途径及网络式调控,过程极其复杂。 MicroRNA(miRNA)是一类存在于真核生物体内,大小为18~25nt的非编码(non-coding)内源性小分子物质,在多种生物学过程,包括细胞增殖、细胞分化、细胞凋亡、以及脂肪的生成代谢中发挥重要作用。选用小鼠、大鼠和鸡胚等实验动物来研究脂肪的生成机制,已经为解决肥胖问题提供了一些依据。但是选用矮小型鸡与正常型鸡来作为实验动物对脂肪的生成机制进行研究还未见报道。为了研究这两个品种鸡之间脂肪生成的差别,我们选用7周龄的矮小型鸡和正常型鸡做为实验动物,运用miRNA微阵列技术分析腹脂中miRNA的表达谱,芯片结果显示:miR-10b,miR-148a,miR-199-3p,let-7k在矮小型鸡中表达上调,正常型鸡表达下调,其中miR-10b达到极显著差异(P﹤0.05);miR-181a-5p,miR-2188在矮小型中表达下调,正常型鸡表达上调,达到显著差异(P﹤0.01)。分析与脂肪生成相关的差异表达的miRNA的功能,能为人类和伴侣动物的肥胖症等能量代谢综合征寻找新的治疗靶标以及调控家畜的脂肪组织发育从而改善肉质提供借鉴和参考。 关键词:鸡 miRNA 脂肪 The Analysis of miRNA Profiles in Abdominal Fat of Dwarf Chicken and Normal Chicken Xia Liang College of Science, South China Agricultural University, Guangzhou 510642,China Abstract:In modern society, obesity has become a global public healthy problem, more and more people pay attention to it. The signaling pathway of fat growth involved in polygene expression, and network control, the process is extremely complex. MicroRNAs (miRNAs) are a class of small non-coding RNAs (approximately 18~25 nucleotides in length), existing in eukaryotic organisms, are involved in the regulatory network of many biological processes and play an important role in many processes ,including cell proliferation, cell differentiation, apoptosis, and fat metabolism. Study the generation mechanism of fat with mice, rats and chicken embryos has provided some basis to solve the problem of obesity. But choose dwarf chicken and normal chicken as experimental animals to study the mechanism of fat formation has not been reported. To investigate the different of fat formation in two types of chicken,we choose 7- week old dwarf chicken and normal chicken as experimental animals. we have adopted miRNA microarray technology to analyze the expression profile of miRNA in abdominal fat.
