Key Laboratory of Environm ent and Genes Related to D iseases, X i an J iaotong U niversity, X i an 710061, China) Abstract: O bjective To investigate the role of adult cardiomyocytes in the differentiation of mesenchymal stem cells (MSCs) into cardiomyocytes. M ethods Rat MSCs were isolated by a Percoll s gradient solution and cultured in low2glucose Dulbecco s modified Eagle s medium (DMEM ). After 2 passages, cell2surface antigen CD34, CD71 and CD90 for rat MSCs were determ ined by flow cytometry, and these MSCs were transfected with pEGFP2N3 by L ipofectam ine2000. Then those MSCs labeled with GFP, were cultured in contacted, nocontacted and conditioned with adult rat myocardiocytes. Immunofluo2 rescence staining against 2actin, desm in, and troponin2T were performed after 1 week. Results Immuno2 fluorescence staining was positive against 2actin, desm in, and troponin2T on MSCs in contacted culture group. In contrast, no 2actin, desm in, and troponin2T exp ression on MSCs were observed in the noncon2 tacted culture group and the conditioned culture group. Conclusion D irect cell2to2cell contact between MSCs and adult cardiomyocytes may induce differentiation of MSCs into cardiomyocytes.
