苦瓜花粉生活力检测方法的比较研究 PAGE  PAGE 16 苦瓜花粉生活力检测方法的比较研究 摘 要 苦瓜是一种重要的蔬菜作物,研究苦瓜花粉活力的检测方法,可以为苦瓜的杂交育种工作提供前期的参考。离体萌发法是普遍认为检测花粉活力最接近花粉的实际生活力,但该法所耗的时间长,所以本研究通过其他方法与离体萌发法比较,选出适宜苦瓜花粉活力快速检测的方法。本研究用碘—碘化钾染色法、TTC染色法、醋酸洋红染色法、过氧化物酶法、无机酸法和离体萌发法对苦瓜花粉活力进行研究,得出碘—碘化钾染色法、TTC染色法、醋酸洋红染色法、无机酸法测出的花粉活力与离体萌发法有极显著的差异,过氧化物酶染色法与离体萌发法测得结果没有达到显著差异,且这两种方法测得苦瓜花粉活性分别为:过氧化物酶染色法04-17×25:62.27%,04-17:70.72%;离体萌发法04-17×25:66.99%,04-17:72.48%。通常认为离体萌发法测出的花粉萌发率是做接近自然状态的花粉活力,所以研究得出的结论为过氧化物酶染色法适宜苦瓜花粉生活力的快速检测。蔗糖基本培养基上添加硼酸能够显著促进苦瓜花粉萌发,苦瓜花粉离体萌发的合适培养基配方和浓度有待进一步研究。 关键词:苦瓜;花粉活力;测定方法 Abstract Bitter gourd is an important vegetable crops, pollen viability of the detection method of bitter gourd, bitter gourd of cross-breeding can provide early information.
