体外细胞单轴循环牵伸载荷模型建立的实验研究
杨广华1,唐康来1,马 林1,周 梅1,陈 万1,穆米多1,袁成松1 ,胡 超1 ,李赛英2 (40038 重庆,第三军医大学西南医院骨科,全军矫形外科中心1;401147 重庆,重庆市机电设计研究院TSCs在培养皿中培养10 h时,全部贴壁且沿微沟槽方向生长。在不同牵伸条件下牵伸模型运行稳定,硅树脂培养皿实际运行频率、应变位移距离和设定频率及理论应变位移距离比较差异无统计学意义(P﹥0.05);TSCs无明显脱落及漂浮;牵伸组F-actin束变细,部分发生断裂,红色荧光(F-actin)染色减弱且分布不均匀。 结论 单轴循环牵伸模型操作简便、参数设计范围宽,可模拟不同条件进行体外贴壁细胞生物力学载荷实验研究。

Experimental study on establishment of a uniaxial cyclic stretching model in vitro

[Abstract] Objective To design a uniaxial cyclic stretching model in vitro. Methods The microgrooved dishes were made of silicone and analyzed strain distributions using the finite element software ABAQUS. TSCs were cultured on dishes and observed adherent growth situation. The mechanical running parts of stretching model cyclically stretched dishes through programmable logic controller (PLC).Dishes were divided into 16 groups when cyclic stretch 24h under different stretch frequency (0.5, 1.0, 2.0, 3.0 Hz) and strain (4%, 8%, 16%, 20%). Silicone dishes actual running frequency and displacement distance were measured; TSCs were observed Falling off and floating using Inverted microscope; F-actin in cytoskeleton were observed using laser confocal scanning microscope under stretching time 8h and 12h (frequecndy 1Hz and strain 8%). Results Silicone dishes strain distribution was uniformity. Effectivestrain range of the bottom membrane accounted for about 91% of its total area. When cultured on dishes 10h, TSCs all attached on dishes and growthed along microgroove direction.
