Abstract:Objective To deve op a simp e, rapid and safe method for extraction of p asmid DNA from
Lactococcus lactis .MethodThe method for p asmid extraction from act ic acid bacteria was improved by combining
the method of E . coli p asmid extraction and the reported method of p asmid extraction from actic acid bacteria
( LAB) . Conditions for this new method were optimized in terms of ysozyme concentrat ion and ysozyme treatment
way and time ength. . Result The opt ima concentration of ysozyme was 10 mg/ m , the opt ima ysozyme
treatment method and time were at 37 for 30 min with shaking. ConclusionThis method avoided the use of
ethidium bromide ( EB) and simp ified the comp icated procedure, so it was a high effective and safe LAB p asmid
DNA extraction method.
