根据前期实验和相关报道,辣木花、叶、籽均含有蛋白酶,且能凝乳,特别是辣木籽的净水功能声名远扬,但未见其深入研究。采用水浸泡、真空浓缩、冷冻干燥以及SDS电泳分离蛋白酶、超滤纯化、ESI质谱酶解肽段α-酪蛋白、-酪蛋白和β-酪蛋白κ-酪蛋白的酶切位点等一级结构变化及产物,揭示辣木蛋白酶的凝乳机理。根据电泳条带、HPLC多肽图谱、FAA等的变化研究辣木籽蛋白酶对污水大分子蛋白质的降解作用;利用扫描电镜、传输扩散波光谱The protease and milk coagulating, water purification properties were found from Morlnga oleifera flower, leaves and seeds according to the former test. Extraction, isolation and identification of the protease from Morlnga oleifera flower, leaves and seeds is going to be developed by the steps of bath, vacuum concentration, freeze-drying, separation, ultrafiltration and mass spectrum. The milk-clotting mechanism of the protease will be researched according to the cleavage site on κ-casein and Casein degradation by SDS,matrix-assisted laser desorption time-of-flight mass and HPLC during coagulation. The water purification mechanism of the protease from Morlnga oleifera seeds is about to be determined according to the sewage protein degradation and the association, flocculating reaction, microscopic structure change by the methods of SDS,HPLC, Confocal Scanning Laser Microscopy, UV spectrum and Diffusing Wave Spectroscopy, et al. The scientific theory of milk-clotting and water purification mechanism from Morlnga oleifera protease is important to develop new chymosin and enzyme engineering purifying agent and benefit to supplement the scientific system of Morlnga oleifera.
