_______________________________________________________________________________ The Dual-coupling Condensor of the Fluorescent 1 Microscope for the Photorelease Technique Li Lan, Anlian Qu, Zhiyong Yang, Yunyan Zhou Institute of Biophysics and Biochemistry, Huazhong University of Science and Technology Wuhan 430074 P.R.China Abstract A novel dual-coupling condenser for the fluorescent microscope was designed and used in some biophysics experiments. Proper glass, silica antireflective coat, carefully correcting of aberrations by software and dichroic beamsplitters were used to maintain optimal throughput and guarantee optimal homogeneity in the specimen plane. The average transmission of condensor is 31.3%. In given conditions, the density of flash energy is 13.85mJ/mm2 in the specimen plane. A result of photorelease experiment done by this condenser is given. It verifies that the condenser is suitable for coupling monochromator and flashlamp in the photorelease application. 1 Introduction Fluorescence illumination and observation is the most rapidly expanding microscopy technique employed today, meanwhile photorelease technique is a very powerful tool for the study of the molecular mechanisms underlying physiological processes[1,2].
