动物科学技术学院：吴军 摘要 本文运用组织学H.E染色法、改良甲苯胺蓝染色法、醛品红—阿利新蓝染色法和阿利新蓝—高碘酸雪夫氏反应染色法对罗非鱼消化道肥大细胞的形态分布等进行研究。 结果发现罗非鱼消化道中存在大量的肥大细胞。肥大细胞大小不一，呈圆形、椭圆形、长梭形、鼓槌形、锥体形或不规则形状，有的可见长突起，细胞核蓝染。肥大细胞主要分布在胃黏膜固有层近黏膜肌层处，粘膜层胃底腺及黏膜肌等结构。体积较小，形态不规则，胞核清晰，胞质内颗粒少。H.E染色，肥大细胞圆或卵圆形，常成群聚集在小血管旁，胞质内充满粗大的嗜碱性颗粒。改良甲苯胺蓝染色，也可显示出一些肥大细胞，胞质颗粒着紫红色。醛品红—阿利新蓝染色，胃肠道的肥大细胞细胞质颗粒呈紫红色至深紫色，弹力纤维也呈深紫色，红细胞呈鲜橙黄色，其余组织为淡黄色。阿利新蓝—高碘酸雪夫氏反应染色，肥大细胞大多数红染，说明胃肠道中含有较多的含中性黏多糖的肥大细胞。 关键词：消化道 肥大细胞 Histochemical Study on Heterogeneity of Mast cells in the tilapia digestive tract Abstract The digestive tract and the forms and distribution of the mast cells in the tilapia were observed by tisssuing, HE staining, improved toluidine blue staining，aldehyde fuchsin-alcian blue staining and alcian blue - xuefushi periodate reaction staining in this paper. The result showed the presence of massive mast cells in digestive tract of tilapia, they were in different sizes and could be round, oval, Long spindle-shaped, drumstick-shaped, cone-shaped or irregular shaped, some may had long process and blue-stained nucleus.Digestive Tract mast cells mainly distribute in the gastric lamina propria near the muscularis mucosa, mucosal layer of the fundic gland mucosa muscle structure. H.E staining, the mast cells circle or round circular, often gather nearby the minute vessel, and within the cytoplasm there were filled with coarse basophilic granules. The improved toluidine blue staining, also demonstrate some mast cells, and cytoplasmic granules into purple. The aldehyde fuchsin-alcian blue staining, cytoplasmic granules of mast cells of the gastrointestinal tract into purple to deep purple, the elastic fibers also assumed into deep purple, other organizations for faint into yellow. alcian blue - xuefushi periodate reaction staining, the mast cell into red, explained that in the gastro-intestinal tract there were many polysaccharide including the neutrality mast cells.
