XXXXXXXX 本科学位论文 12-去氧佛波醇-13棕榈酸酯和没食子酸乙酯对乳腺癌细胞增殖的影响 The effect of 12-deoxyphorbol-13-palmitate and ethyl gallate on the proliferation of breast cancer cells 学 生 姓 名： 院 （系）： 学 生 年 级： 专 业 名 称： 指导教师姓名： 指导教师单位： 协助指导教师： 协助导师单位： 论文提交日期：2014 年 6 月 16 日 摘要 目的：探讨狼毒大戟中两种单体药物12-去氧佛波醇-13-棕榈酸酯（12-Deoxyphorbol-13-Palmitate,12D-13P)和没食子酸乙酯（EG）对乳腺癌细胞MDA-MB-231增殖的影响。方法：采用MTT法检测两种单体药物12D-13P和EG对乳腺癌细胞MDA-MB-231增殖的影响，用二甲基亚砜溶解两种药物，并设置无药对照、溶剂对照组、以及5μg/ml、10μg/ml、20μg/ml、40μg/ml和80μg/ml浓度的药物组，37℃恒温箱中培养24h后，加入MTT孵育4h后，弃去上清液加入二甲基亚砜，用酶标仪在490nm的波长处测定OD值，计算细胞生长抑制率。结果：与对照组相比，12D-13P作用24h、48h后，10、20、40、80μg/ml剂量组明显抑制乳腺癌细胞的生长，差异有显著性(P0.05)；EG作用24h、48h后10、20、40、80μg/ml剂量组明显抑制乳腺癌细胞的生长，差异有显著性(P0.01)。结论：狼毒大戟中12D-13P和EG两种单体药物对乳腺癌细胞MDA-MB231的增殖有显著性抑制作用。 关键词：狼毒大戟；12D-13P；EG；MTT Abstract Objective: To investigate the effect of 12-deoxyphorbol-13-palmitate (12D-13P) and ethyl gallate (EG), which two kind of monomer come form euphorbia fischeriana, on the proliferation of breast cancer cell. Methods: Using the MTT assay the inhibition of 12-deoxyphorbol-13-palmitate and ethyl gallate on breast cancer cells. After dissolving 12D-13P and EG in dimethy sulfoxide, the cell were divided into control group, experimental group(5,10,20,40,80μg/ml) and solvent control group, each group set up three samples. To make breast cancer cells incubated at 37℃ temperature for 24h and 48h. After that, adding MTT and contiue incubateding for 4h, then the liquid were discarded and add dimethy sulfoxide. Detecting the absorbance value with microplate reader on the wavelength 490nm, then the average inhibition rate can be obtained. Results: MTT assay showed after 24h and 48h, the12D-13P group compare with control group, the concentration(10,20,40,80μg/ml) of 12D-13P significantly inhibit the cell MDB-MA-231 proliferation (P0.05). In contrast to the control group the EG group the concentration(10,20,40,80μg/ml) of EG significantly inhibit the cell MDB-MA-231 proliferation (P0.01). Conclusion: 12-de
