不同植物生长物质诱导洋桔梗愈伤组织的研究 摘要:洋桔梗是国际上十分流行的盆花和切花种类之一。开展洋桔梗愈伤组织诱导的研究,对于洋桔梗的纯合多倍体诱导及新种质的培育等均具有重要意义。本研究以洋桔梗‘Double Mariachi Pink’品种的无菌苗叶片为外植体,以MS为基本培养基,附加不同浓度配比的6- BA、TDZ和NAA、IBA,研究其对洋桔梗愈伤组织诱导的影响。结果表明,单独使用植物生长物质出愈率和愈伤组织生物量明显低于植物生长物质组合时的结果。各种植物生长物质组合均能诱导出愈伤组织,且出愈率高,很多培养基的外植体出愈率达100%,只是产生愈伤组织生物量的多少不一,很多诱导培养基还诱导出了不定根和不定芽。6-BA与NAA的不同浓度配比诱导外植体产生的愈伤组织生物量最多,诱导出不定根和不定芽的组合最多;6-BA与IBA的不同浓度配比诱导产生愈伤组织的时间最长且愈伤组织生物量最少,诱导出不定根和不定芽的组合也最少;不同浓度的TDZ与IBA组合诱导产生愈伤组织的时间最短。综合考虑出愈率、出愈时间、愈伤组织生物量及鲜重与干重,培养基MS+7.5μM TDZ+0.1μM IBA每一项结果均最优,故最佳的愈伤组织诱导培养基为MS+7.5μM TDZ+0.1μM IBA。 关键词:洋桔梗;叶片;植物生长物质;愈伤组织;诱导 Study of Different Plant Growth Substances Induction Callus of Eustoma grandiflorum Abstract : Eustoma grandiflorum is one type of the popular potted flowers and cut flowers in the international. Study on callus induction of Eustoma grandiflorum is of great significance for the homozygous polyploid induction and the breeding of new varieties of Eustoma grandiflorum. In this study, aseptic seedling leaf of ‘Double Mariachi Pink’ varieties as explant, MS as the basic culture medium, supplemented with different concentration ratio of 6 - BA, TDZ and NAA, IBA, callus induction of Eustoma grandiflorum was studied. The results showed that Single-use plant growth substances?of Callus induction rate and?callus biomass were significantly lower than the results of the combinations of plant growth substances. A variety of combinations of plant growth substances could induce callus, and the rate was high, a lot of explants on the culture medium of callus rate reached 100%, just callus biomass was not the same, many mediums also induced adventitious roots and adventitious bud. Different concentrations of 6-BA and NAA induced explants produced the most Callus biomass, adventitious root and adventitious bud; different concentration ratio of 6-BA and IBA took the longest time to induce callus formation and callus biomass was the least, and the least combination induced adventitious root and adventitious bud; different conc
