Sci .2007 ,35( 30) :9464 ,9469 责任编辑 罗芸 责任校对 李菲菲 一种在光镜下观察花粉滚动和立体形状的制片方法 洪亚平, 薛娴, 段春燕 ( 河 南科 技大学 农学 院, 河南洛 阳471003) 摘要 介 绍利用油镜 油观察花粉滚动和立体形状的制 片方法, 以及将 普通显微镜( 即明场显微镜) 改制成 暗场显微 镜 的方法和 观察结 果。将植物花粉均匀地撒在载玻片上, 盖上盖玻片, 用铅笔上的橡皮头等软物轻敲1 ～2 下, 然后在盖玻片外侧( 或角端) 滴上 1 小滴油镜 油, 油镜 油沿着载玻片与盖玻片之 间的缝隙向内扩散, 制片完成并 立即进行 显微镜观察 。利用 油镜 油对 花粉进行制 片, 比较容 易在40 倍物镜下直观地观察和记录花粉 的滚动过程和立体形状、层次结构、表 面纹饰 。将显微镜进行一些结构改建, 可 以一镜多用。随着光学 显微镜的不断升级换代, 光镜在观察花粉形状和结构信 息等方面, 将具有越来越多电镜无法取代的优势。 关键词 光学显微镜; 花粉; 滚动; 立体形状; 制片 中图分类号 Q944 .42 文献标识码 A 文章编号 0517 - 6611( 2007) 30 - 09464 - 01 A Flaking Method of Observing the Rolling and Stereo Shapes of Pollen under Optical Microscope HONG Ya- ping et al ( College of Agronomy , Henan University of Science and Technology , Luoyang , Henan 471003) Abstract The study ai med to introduce a flaki ng method of usi ng the i mmersion oil to observe the rolli ng and stereo shape of pollen , as well as the method of reorganizing common microscope ( bright field microscope) into dark field microscope and the observation result . The microscope observation should be conducted at once after the flaking was finished by sprayi ng botanic pollen on the microscope glass slide , covering the cover glass and knocking it for 1 ～2 ti mes with soft things such as eraser onthe pencil , then dropping a small drop of i mmersion oil on the outside or corner of cover slip , with the oil diffusing along the gap between microscope glass slide and cover glass into inside .The rolling course , stereo shape , hierarchy structure and surface or- namentation of pollen were easier to observe and record visually under 40-fold microscope by flaking pollen with i mmersion oil .
