5-氟尿嘧啶及长春新碱对胃癌细胞SGC-7901干细胞相关特性的影响 陈 军,余佩武 (400038 重庆,第三军医大学西南医院全军普通外科中心,微创胃肠外科中心R735.2 [文献标志码] A Effects of 5-FU and vincristine on gastric cancer stem cells-associated phenotypes and properties in SGC-7901 Chen Jun,Yu Peiwu(General Surgery Center of PLA, Southwest Hospital, Third Military Medical University, Chongqing, 400038, China ) [Abstract] Objective To identify the effects of 5-FU and vincristine (VCR) on gastric cancer stem cells-associated phenotypes and properties in gastric cancer cell line SGC-7901. Methods SGC-7901 were treated with 5-FU and VCR respectively. Candidate gastric cancer stem cells’ marker CD44 and CD133 were assayed as well as other stemness genes and EMT associated genes. Besides, some assays such as colony formation, sphere formation and tumorigenic formation were used to test the properties associated with cancer stem cells. Results After 5-FU and VCR treatment, the mRNA expression levels of CD44 and CD133 were more in all of the doses in current experiment (up to 2.36±0.03, 1.57±0.07, 6.03±0.7, 4.06±0.76 folds respectively, P<0.05). But the capability of colony formation and tumorigenic formation didn’t increased. However, after VCR treatment SGC-7901 manifested the EMT gene profile and showed different morphologic features in serum-free culture, which was different from 5-FU treatment. Conclusion Transient treatment of 5-FU and VCR respective promoted the expression of CD44 and CD133. Besides, VCR induced EMT changes of SGC-7901 and enhanced sphere formation in serum-free culture medium.
