肝细胞癌中FoxM1与AFP转录及分泌的相关性研究 艾剑刚，陈娟娟，董玉芳，黄世峰，王海霞，唐翌姝，张莉萍（400016 重庆，重庆医科大学附属第一医院检验科） [摘要] 目的 肝细胞癌（hepatocellular carcinoma,HCC）中FoxM1与AFP转录及分泌的关系。 方法 采用Western blot和荧光定量PCR技术检测例HCC癌组织中FoxM1蛋白和AFP mRNA表达水平，电化学发光法检测血清AFP分泌水平，分析三者相关性；采用RT-PCR和Western blot技术检测FoxM1特异性抑制剂硫链丝菌素（thiostrepton，TST）作用人肝癌HepG2细胞和HepG2.2.15细胞24小时和48小时后FoxM1 mRNA和蛋白表达水平变化，电化学发光法检测培养基上清AFP分泌水平变化。 结果 HCC癌组织中FoxM1蛋白表达水平与AFP mRNA表达水平（r=0.448,P﹤0.01）和血清分泌水平（r=0.381,P﹤0.01）呈正相关，AFP mRNA与血清中AFP分泌呈显著正相关（r=0.618,P﹤0.01）；与未处理组相比，TST作用HepG2细胞和HepG2.2.15细胞24小时和48小时后FoxM1 mRNA和蛋白表达水平及培养基上清AFP分泌水平均明显下降（P﹤0.001）。 结论 肝细胞癌中FoxM1与AFP转录及分泌水平密切相关，FoxM1在 AFP的表达调控中可能发挥重要的调控作用。 [关键词] 肝细胞癌；FoxM1；AFP mRNA；AFP[基金项目] 国家自然科学基3097337881272545）；重庆市自然科学基金项目（No.2010BB5390）；国家临床重点专科建设项目基金；重庆市卫生局课题（2010-2-090） [通信作者] 张莉萍 Tel:(86)023-mail:Liuzhangcq@ Correlation of FoxM1 and AFP in hepatocellular carcinoma Ai Jiangang, Chen Juanjuan, Dong Yufang, Huang Shifeng, Wang Haixia , Tang Yishu, Zhang Liping (Departiment of Medical Laboratory, First affiliated Hospital, Chongqing Medical Univercity, Chongqing, 400016, China) [Abstract] Objective To investigate the relationship between forkhead box M1(FoxM1) and Alpha-fetoprotein(AFP) in hepatocellular carcinoma(HCC). Method The expression of FoxM1 and AFP mRNA in 52 HCC tissues were detected by Western blotting and qRT-PCR，and AFP in serum was detected by electrochemiluminescence method.The correlation among the expression of FoxM1,AFP mRNA and serum AFP were analyzed.
