刺参(Apostichopus japonicus)肠道潜在益生菌对稚参生长、免疫及抗病力的影响
摘要:16S rDNA序列法鉴定了3株分离自刺参肠道的潜在益生菌分别为Bacillus sp.(GSC-1),Bacillus sp.(GSC-2) 和Enterococcus sp.(GSC-3)。通过浸浴实验分别检验了3株菌对稚参期刺参的安全性,(加上安全性检验的结果)然后将(具体菌株)和鼠尾藻粉以109 CFU/g的比例混合后饲养稚参20d,结果表明GSC-1和GSC-3均能提高稚参的成活率特定生长率和变色率,GSC-1显著提高了稚参的成活率和变色率(P0.05)特定生长率(P0.05);GSC-1可有效降低稚参弧菌数(P0.05)GSC-1和GSC-3均可以提高稚参对灿烂弧菌的抵抗力,其中GSC-3显著降低了灿烂弧菌攻毒后稚参的死亡率(P0.05)。本GSC-1 和GSC-3,提高了抗病能力,因而GSC-1和GSC-3可作为益生菌应用于稚参养殖中。

Abstract
Three strains of potential probiotics (GSC-1, GSC-2, GSC-3), isolated from the intestines of Apostichopus japonicus, were identified by 16S rDNA sequences genotyping. Their toxicity to juvenile sea cucumber (average weight 0.05g) was evaluated. Then an experiment was conducted to investigate the effects of these potential probiotics on the growth performance and innate immune response of A. japonicus. The potential probiotics were identified as Bacillus (GSC-1), Bacillus (GSC-2) and Enterococcus (GSC-3), respectively. The toxicity study showed that GSC-1 and GSC-3 were safe for juvenile sea cucumber while GSC-2 caused higher mortality at 107 CFU/ml. A 20-day feeding trial was conducted. Feeds were algae (Sargassum thunbergii) powder supplemented with GSC-1 or GSC-3 at 0 (control) and 109 CFU /g respectively. SGR and survival rate were significantly increased in treatment supplemented with GSC-1 while survival rate and discoloration rate were remarkably improved in treatment of GSC-3 (P0.05). Moreover, probiotic GSC-1 increased total bacteria counts and decreased Vibrio spp. counts significantly (P0.05). Both GSC-1 and GSC-3 produced positive effects on sea cucumber immune responses and disease resistance, especially GSC-3 had significant improvement in the disease resistance after the Vibrio splendidus exposure (P0.05).
