The medium mainly contained pig litter, which was produced from microbial fermentation bed for raising pigs, and the sporulation yield was used as the indicator in all tests.. The results of single factor tests showed that the ratio of pig litter and wheat bran, the content of wheat grain, carbon and nitrogen were 9:1, 40.00% of wheat grain, 3.00% of sucrose, and 2.00% of ammonium sulfate, respectively. The results showed that wheat grain, sucrose and ammonium sulfate were significant for the spore of Trichoderma harzianum based on the response surface methodology analysis(P0.05).The optimum medium constitutes were as followed: 9:1 of bedding of microbial pig raising and wheat bran,wheat grain 36.74%, sucrose 3.07%, ammonium sulfate 1.75%, and the average yield of spore reached to 2.98×109cfu·g-1 ,.which was closed to the actual fermentation.
