HBV感染者血清及PBMC培养上清液IL-12水平检测及意义【摘要】目的：探讨急性乙型肝炎、慢性乙型肝炎、HBsAg携带者及正常人血清和PBMC培养上清液中IL-12水平及其意义。方法：应用Ficoll Hypaque密度梯度离心法从肝素化外周血中分离PBMC，用ELISA方法检测血清及PBMC培养上清液IL-12水平。结果：（1）血清IL-12：急性乙型肝炎血清IL-12阳性率30%，慢性乙型肝炎50%，HBsAg携带者10%，正常人为0。（2）PBMC培养上清液IL-12水平：急性乙型肝炎（121.09±97.43）pg/ml，慢性乙型肝炎（57.11±40.51）pg/ml，HBsAg携带者（25.4±15.86）pg/ml，正常人（38.35±33.86）pg/ml。结论：慢性乙型肝炎血清IL-12阳性率显著高于正常人，急性及慢性乙型肝炎PBMC培养上清液IL-12水平显著高于HBsAg携带者，提示HBV感染者血清IL-12阳性率及PBMC培养上清液IL-12水平与肝脏炎症活动有关。 【关键词】乙型肝炎；HBsAg携带者；PBMC；IL-12 文章编号：1009-5519（2007）22-3343-02 中图分类号：R446 文献标识码：A IL-12 levels of the serum and PBMC culture supernatant in patiants with HBV infection and its significance JIANG Feng-peng1，YU Xiao-dong2，HUANG Fen3 （1.Anshan Infectious Diseases Hospital，Anshan 114011，China；2.The Second Hospital of Anshan，Anshan 114012，China；3.The Second Affiliated Hospital of China Medical University，110003，China） 【Abstract】Objective：To detect the IL-12 levels of the serum and PBMC culture supernatant in patients with acute hepatitis B，chronic hepatitis B，HBsAgcarriers and healthy persons and discuss its significance.Methods：PBMC was isolated by centrifugation on a Ficoll Hypaque density gradient.The IL-12 levels of the serum and PBMC culture supernatant were detected by ELISA method.Results：（1）Serum IL-12：The serum IL-12 positive rate in patients with acute hepatitis B，chronic hepatitis B，HBsAgcarriers and healthy persons was 30%，50%，10%，0 respectively.（2）PBMC culture supernatant IL-12:The IL-12 levels of PBMC culture supernatant in patients with acute hepatitis B， chronic hepatitis B，HBsAg carriers and healthy persons were （121.09±97.43）pg/ml，（57.11±40.51）pg/ml，（25.4±15.86）pg/ml，（38.35±33.86）pg/ml respectively.Conclusion：The serum IL-12 positive rate in patients with chronic hepatitis B was significantly higher than that in healthy persons.The IL-12 levels of PBMC culture supernatant in patients with acute hepatitis B and chronic hepatitis B were significantly higher than those in HBsAgcarriers.The IL-12
