人脐血源基质细胞对造血细胞归巢植入能力的影响 刘 颖,陈幸华,张 曦,高 蕾,张 诚,冯一梅(400037重庆,第三军医大学新桥医院血液科) [摘要] 目的 探讨人脐血源基质细胞(hUCBDSCs)对造血细胞早期归巢及植入能力的影响。 方法 CCK-8法和Transwell法分别比较hUCBDSCs和人骨髓基质细胞(hBMSCs)对脐血单个核细胞(hUCB-MNCs)体外黏附及迁移能力的影响;RT-PCR法检测hUCBDSCs对促归巢相关因子的表达;60Coγ射线致死剂量辐照BABL/c小鼠后分别接受hUCB-MNCs单移植或hUCB-MNCs联合基质细胞共移植,流式细胞仪检测小鼠骨髓人源CD45+细胞植入率;追踪移植后CM-DiI标记的hUCB-MNCs体内迁移情况,并比较各组归巢率。结果hUCBDSCs和hBMSCs均能促进hUCB-MNCs的黏附,且迁移作用显著强于无基质细胞共培养对照(P0.05);hUCBDSCs能表达一系列与造血归巢相关的黏附分子和细胞因子;hUCBDSCs在联合hUCB-MNCs共移植中能不同程度提高造血植入率,且当输注的hUCB-MNCs为低剂量(两种细胞按1:1各移植2×106个/只)R551 [文献标志码] A [基金项目]国家自然科学基金 ();重庆市自然科学基金重点资助项目(CSTC2009BA5011) [通信作者]陈幸华, E-mailxhchen888@ Influence of hUCBDSCs on homing efficiency and engraftment capability of hematopoietic cells Liu Ying,Chen Xinghua,Zhang Xi,Gao Lei,Zhang ChengFeng Yimei (Department of Hematology, Second Affiliated Hospital, Third Military Medical University, Chongqing 400037 , China) [Abstract] Objective  To observe the effect of hUCBDSCs on homing efficiency and engraftment capability of hematopoietic cells. Methods The effect of hUCBDSCs and hBMSCs on adhesion and migration capability of hematopoietic cells in vitro were measured by CCK-8 and Transwell assay, respectively. RT-PCR was applied to detect the homing-related factors expressed by hUCBDSCs. Additionally, the lethally irradiated (60Coγ ray) BABL/c mice were transplantated with hUCB-MNCs alone, or combined with stromal cells, respectively. The engraftment rates of human CD45+ cells were determined by flow cytometry. The homing efficiency of hUCB-MNCs labeled by CM-DiI were observed by laser scanning confocal microscope, and the percentages of homing in each group were compared. Results hUCBDSCs和hBMSCs could promote the adhesion of hUCB-MNCs, and the migration rates of hUCB-MNCs were significantly higher (P0.05) when co-cultured with these two stromal cells.
