白藜芦醇纳米乳对人肝癌细胞系HepG2 生长的抑制作用 杨宝平 欧阳五庆* (西北农林科技大学 动物科技学院,陕西 杨陵 712100)检测纳米乳的形态及粒度分布16.5±1.5nm,分布在10~40nm;经4mg/L Res原料药和4mg/L Res纳米乳处理人肝癌细胞系HepG2 36h 后,与对照组比较细胞增殖水平都显著降低 (p0.01),细胞出现典型的凋亡状态(我觉得应该具体描述什么状态);白藜芦醇纳米乳与白藜芦醇原料药相比,当白藜芦醇浓度大于8mg/L时,细胞抑制率显著增加,有统计学意义(p0.05),IC50分别为:6.5mg/L、24.2mg/L。结论:Res纳米乳对体外培养的人肝癌细胞系HepG2的细胞抑制率高于Res原料药,两者存在显著性差异。 [关键词] 白藜芦醇;纳米乳;细胞凋亡;HepG2 Depressant effect of resveratrol Nanoemulsion on human heptoma cell line HepG2 cells YANG Bao-ping OUYANG Wu-qing* LIU Yu-mei MA Shu-yan (College of Animal Science and Technology, Northwest of AF University, Yangling Shaanxi 712100,China) ABSTRACT: Objective: Resveratrol nanoemulsion was preparated and compare depressant effect on human heptoma cell line HepG2 cell with resveratrol. Methods: Prepared resveratrol nanoemulsion with phase transformation emulsification and tested its shape and particle size. MTT assay was used to observe the effects of resvertrol namoemusion on HepG2 cell growth, and the growth curve was detected. The variation of cell and HE staining revealed abnormal morphology in HepG2 cells treated with resvertrol namoemusion. Results: The shape of resveratrol nanoemulsion was sphere under TEM, with a mean diameter was 16.5±1.5nm, distributing between10~40nm. After treatment with resvertrol namoemusion and resvertrol for 24h respectively, cell proliferation degraded significantly (p0.01), and cells appeared apoptotic characteristic, compared with control group. When the concentration of resvertrol was higher than 8mg/L, the rate of cell inhibition with resveratrol nanoemulsion was significant increased compared with control group, and IC50 were 6.5mg/L,24.2mg/L respectively. Conclusion: The rate of cell inhibition on human heptoma cell line HepG2 cell with resveratrol nanoemulsion was significant higher than resveratrol.
