[摘要]目的观察瘦素对糖尿病性心肌病(diabetic cardiomyopathyDCM)大鼠心室重构的影响。方法10只SPF级健康SD大鼠8周龄170~190g)作为正常对照组(NC组),予以普通饲料喂养。另0 SPF级健康SD大鼠8周龄170~190g)采用高糖高脂喂养后2次腹腔注射链脲佐菌素(STZ)的方法建立DCM大鼠模型,将DCM大鼠随机分为A组、瘦素50 μg/kg组(B组)、瘦素100 μg/kg(C组)。在第11周时,B组、C组分别给予每日腹腔注射瘦素50、100 μg/kg,而NC组与A组均注射等量蒸馏水,持续5第12周末检测组空腹血糖、胰岛素、瘦素及血管紧张素Ⅱ(Angiotensin Ⅱ,AngⅡ)水平;心脏彩超测定大鼠心脏左室舒张末期内径(left ventricular internal diameter at end-diastoleLVIDd)、左室舒张末压(left ventricular end-diastolic pressureLVEDP)、左室射血分数(left ventricular ejection fractionLVEF)。结果与NC组相比,A组出现心功能减退,其瘦素、胰岛素及AngⅡ水平升高;不同剂量瘦素干预后,B组与C组大鼠心功能各项指标变化显著,心功能减退更为严重,瘦素、胰岛素及AngⅡ水平显著升高,且C组变化程度显著高于B组。结论瘦素可能通过神经内分泌系统参与了糖尿病心肌病的发生发展,且与胰岛素抵抗、肾素-血管紧张素系统等密切相关。[关键词] 瘦素;糖尿病心肌病;胰岛素抵抗;血管紧张素Ⅱ
Effects of Leptin on Ventricular Remodeling in Diabetic Cardiomyopathy Rats

Abstract]Objective To establish the diabetic cardiomyopathy rat model, thenobserve the effects of leptin on ventricular remodeling in diabetic cardiomyopathy rats. Methods Ten healthy male SD rats (SPF, 8 weeks,weighing 170 ~ 190g) wereselected as normal control group (NC), those who were given normal diet. Then other fifty healthy SD rats (SPF, 8 weeks,weighing 170 ~ 190g) were intraperitoneally injected twicewith streptozocin(STZ) to induce diabetic cardiomyopathy. The diabetic cardiomyopathy rats were randomly divided into three groups:the DCM group(n=10the 50μg/kg leptin group(n=10) and the 100μg/kg leptin group(n=10). The diabetic cardiomyopathy rats were fed on high fat/high sucrose diet. In the eleventh weeks, the latter two groups were respectively given daily intraperioneal injections of leptin on 50μg/kg and 100μg/kg, while the NC and A groups were injected with distilled water for 5 days.Serum IL-6,Ang-Ⅱand LVEDP,LVIDd, LVEF were analysed after 12 weeks later.ResultsThere were abnormal cardiac function in DCM rats.
