决明子提取物对急性肝损伤保护作用实验探究  【关键词】 提取物 摘要:目的观察决明子提取物对CCl4所致小鼠急性肝损伤的 影响 。 方法 60只雄性昆明小鼠随机分为6组:正常对照组、模型对照组、阳性对照组、大、中、小剂量给药组。大、中、小剂量给药组按21.6,10.3,5.1 g/kg灌胃决明子提取物,阳性对照组按0.2 g/kg灌胃联苯双酯,正常对照组及模型对照组以等体积生理盐水灌胃,共7 d。第6天,模型对照组、阳性对照组和给药组按2.0 ml/kg背部皮下注射CCl4,正常对照组背部皮下注射等体积生理盐水。比较各组血清门冬氨酸氨基转移酶(aspartate transaminase,AST)、丙氨酸氨基转移酶(alanine transaminase,ALT),超氧化物歧化酶(superoxide dismutase,SOD)、丙二醛(malondialdehyde,MDA),并观察肝组织形态学变化。结果决明子提取物能显著降低CCl4所致小鼠血清AST 、ALT含量的升高(Plt;0.01),并能升高SOD的活性,降低MDA含量(Plt;0.05);减轻对肝细胞的病理性损害。结论决明子提取物对CCl4所致的小鼠急性肝损伤具有一定的保护作用。 关键词:决明子; 急性肝损伤; 保护作用 Experimental Study on Protective Effect of Semen Cassiae Extract Against Acute Liver Injury Abstract:ObjectiveTo observe the effect of Semen Cassiaeextract against carbon tetrachloride induced actute liver injury in mice. MethodsSixty male mice were randomly divided into normal control group,model control group,bifendate pills control group,high-dosage group,middle- dosage group and low- dosage group. The mice in the above groups were fed for 7 days. The drug groups were pretreated intragastrically with 21.6,10.3,5.1 g/kg extract,bifendate pills groups were pretreated intragastrically with 0.2 g/kg bifendate. Control animals were given the same volumn physiological saline. On the 6th day of feeding, the mice except normal control group were given 2.0 ml/kg carbon tetrachloride by subcutaneous injection. Twenty-four hours after the intoxication,the serum AST,ALT,SOD activities and MDA content were measured in each group above. ResultsSemen Cassiae extract significantly decreased carbon tetrachloride induced actue hepatic injury with increasing the activities of AST,ALT and the content of MDA ,increased the activities of SOD in serum,the severity of liver injury was slight in the high dosage group in pathological examination. ConclusionIt is revealed that Semen Cassiae extract could prevent carbon tetrachloride from inducing acute liver injury. Key words:Semen Cassiae extract; Liver injury; Protective eff
