一株中度嗜盐蜡样芽孢杆菌分离鉴定和其对苯胺降解特性   摘要:从江苏省连云港市某化工染料厂排污管道的污水中驯化、分离得到1 株能以苯胺为惟一碳源的降解菌株HYHB-3,经形态学特征及生理生化特性观察,结合16S rRNA基因序列分析,初步鉴定为蜡样芽孢杆菌(Bacillus cereus),并研究其对苯胺的降解特性和影响因素。结果表明,菌株HYHB-3 为该菌株属中度嗜盐,最佳生长温度25 ℃,超过35 ℃停止生长;苯胺浓度100 mg/L是HYHB-3菌株降解最适浓度,降解率达77.9%,250 mg/L为饱和浓度; 随时间延长降解效率有增加趋势;菌株HYHB-3降解苯胺的最适温度是25 ℃,降解率达69.9%,且对低温条件具有一定的适应性,15~20 ℃条件下均有一定的降解;降解苯胺最适pH为8,降解率达到最大值,达58.4%;在NaCl含量1%~9%的范围内均有一定的降解,3%、5%时降解率均超过50%以上,9%时仍然有11.7%的降解率。苯胺浓度250 mg/L以上则可以抑制该菌的生长。 关键词:蜡样芽孢杆菌(Bacillus cereus);分离鉴定;苯胺;降解 中图分类号:S852.61+6 文献标识码:A 文章编号:0439-8114(2014)02-0308-04 Isolation and Identification of a Moderately Halophilic Bacillus cereus and Its Degradation Characteristic on Aniline WANG Hong-bin1,2,SONG Xiu-mei1,ZHENG Jin-long1,YAN Bin-lun2 (1.Marine College, HuaiHai Institute of Technology, Lianyungang 222005,Jiangsu,China; 2. Jiangsu Key Laboratory of Marine Biotechnology, Lianyungang 222005, Jiangsu,China) Abstract: A highly efficient degradation bacterium HYHB-3 with aniline as its sole carbon source was isolated from the sewage pipe of a chemical dye factory in Lianyungang, Jiangsu Province. According to the morphological, physiological and biochemical characteristics, combined with 16S rRNA sequence analysis, it was identified as Bacillus cereus. Its degradation characteristics on aniline and factors influencing the degradation were studied. The results showed that the strain HYHB-3 was moderately halophilic with optimum growth temperature of 25 ℃. It might stop growing over 35 ℃. The aniline mass concentration of 100 mg/L was the optimal degradation concentration of strain HYHB-3, with the highest degradation rate 77.9%. 250 mg/L was the saturation concentration. The optimum degradation time was 96~120 h. The degradation efficiency increased with time. The optimum temperature of aniline degradation of Strain HYHB-3 was 25 ℃ with degradation rate 69.9%.
