车前子化学成分的研究 STUDIES ON THE CHEMICAL CONSTILUENTS OF THE SEEDS FROM PLANTAGO DEPRESSA WILLD. 专 业：食品科学与工程 姓 名： 指导教师姓名： 申请学位级别：学 士 论文提交日期：2008年6月10日 学位授予单位：天津科技大学 车前子为车前科植物车前或平车前的干燥成熟种子，含有较多的药用活性成分，具有清热利尿，渗湿通淋，明目祛痰的功能，用于治疗水肿胀满，暑湿泄泻，目赤肿痛，痰热咳嗽等症状。车前子中的药用有效成分可用于功能性食品的开发。本文对车前子（平车前）进行了分离提取，并对所提取的化学成分进行了初步研究。 用60%乙醇对车前子进行浸提，得到粗提取物，再通过大孔树脂柱层析，水-95%乙醇进行梯度洗脱，将提取液进行初步分离。将50%乙醇洗脱部分通过硅胶柱层析和ODS柱层析结合高效液相色谱、薄层色谱进行进一步分离纯化，现分离出四种化学成分，通过对分离的化学成分进行结构鉴定，其中两种物质已经确定结构，其化学结构分别确定为车前子苷(Plantagoside ，1 、Dihydrotricetin( 2 )。其中Dihydrotricetin是初次从此植物中分离得到。 大量制备车前子苷标准品，纯度达98%以上用以上车前子苷标准品制备标准曲线，建立车前子苷的高效液相色谱检测方法，并对几个样品进行了含量测定。 Dihydrotricetin ABSTRACT Semen Plantaginis is the dry and matuer seeds of Plantaginaceae including Plantago asiatica L. and Plantago depressa Willd. which contains many kinds of medical elements. It is effective inreducing fever,causing diuresis,improving acuity of vision,and expectorative effect. It is used to treat edema, summerheat, diarrhea, cough with phlegm, red eyes and sore throat. The medical elements of Semen Plantaginis can be used to functional food development. In the paper, the separation and the chemical composition of the seeds of Plantago.depressa Willd. were reported. The seeds of Plantago depressa Willd. are was extracted by 60%ethanol solution, followed by macroporous resin column, gradient eluting with water-95% ethanol. The 50% ethanol moiety is purfied by successive column chromatography on silica gel and ODS with HPLC and TLC method. Now four mechical compositions have been isolated, the structure of which are indentified with ESI-MS , 1H NMR and 13C NMR spectral. Two of them are determined. The structure of them are respectively Plantagoside(1), Dihydrotricetin (2) . Prepare Plantagoside standard(degree of purity is above 98%). Build HPLC test method by Preraring standard curve with Plantagoside standard,and measure the content of Plantagoside in sample. Keywords: Semen Plantagini
