And in order to obtain valid data, cell lines for the study need to be carefully and correctly selected. In recent years, many cases about cross-contaminated or misidentified cell lines have been reported. A contaminated cell line would not only render the data invalid, their distribution around the labs worldwide can also cause tremendous damage to the future research. We accidentally found surprisingly strong hybridization signals appeared when mouse genomic DNA probe was hybridized to the DNA sample of the human SH-SY5Y cell line of our lab, suggesting that this cell line may be contaminated with mouse cells. To investigate and identify the possible contamination and its range, we used various techniques including PCR, genomic clones sequencing, chromosome counting, southern dot blot and STR fingerprinting analysis to analyze samples from our lab and six other laboratories. The data suggested that some SH-SY5Y cell lines were indeed contaminated with mouse cells. In samples from seven labs, there were only three authentic SH-SY5Y cell lines, while the other four contained mouse cells substantially or indeed were mouse cells completely. Our study suggested that mouse cell contamination in SH-SY5Y cell line need to be taken into consideration seriously and DNA hybridization combined with clone sequencing may be an economical strategy in cross species contamination identification.
