[摘要]目的:研究姜黄素对A549细胞增殖的抑制作用,探讨其分子机制。方法:分别用平板克隆实验、MTT检测不同浓度姜黄素对A549细胞克隆形成数和增殖的影响。于不同时间点,用FCM检测细胞周期;RT-PCR、Western-Blot检测P53、P21 、PCNA及eIF4E基因的表达。结果:姜黄素可引起细胞S期阻滞,抑制细胞增殖,减少细胞的克隆形成数。姜黄素作用A549 24h后P53 mRNA表达上调,48h后P21 mRNA、P53和P21蛋白表达上调,eIF4E mRNA、PCNA mRNA和蛋白表达下调,72h后eIF4E蛋白表达下调,较对照(DMSO)组,差异有统计学意义(P0.05)。结论:姜黄素可减少A549的克隆数、阻滞细胞于S期、抑制细胞增殖,其机制可能与姜黄素促进P53和P21表达,抑制PCNA和eIF4E表达有关。

Inhibitory Effect of Curcumin on Proliferation in Lung Cancer A549 Cells

[ABSTRACT] Objective: To investigate the inhibitory effect of curcumin on proliferation in A549 cells and explore the possible mechanisms. Methods: A549 cells were treated with curcumin at different concentrations or time intervals as indicated, after which clone formation and proliferation were determined by plate cloning and MTT, respectively. The cell cycle was measured by flow cytometry. The expression of P53, P21, PCNA and eIF4E at mRNA and protein levels were detected by RT-PCR and Western-Blot. Result: Exposure of cells to curcumin resulted in cell cycle arrest at S phase, inhibition of cell proliferation, and reduction of colon formation. Treatment of cells with 20 μmol/L curcumin for 24 h also resulted in increase in expression of p53 and p21 at mRNA and protein levels, these events became apparent after 48 h or 72 h of drug exposure. In addition, exposure of cells to 20 μmol/L curcumin for 48 h or 72 h resulted in decrease in the expression of eIF4E and PCNA at mRNA and protein levels.
