毕业设计(论文) (2009届) 题 目 脂肪醇聚氧乙醚降解菌的 筛选和鉴定 摘要 脂肪醇聚氧乙烯醚降解菌的筛选和鉴定 摘 要:随着现代生物技术的发展,微生物在环境保护方面的应用前景越来越广阔。本研究通过用筛选脂肪醇聚氧乙烯醚(alkyl alcohol polyoxyethylene ether, AEO)降解菌的方法,对我国工业中用量最大的非离子表面活性剂AEO进行降解特性研究。 从常州市清潭污水处理厂采集活性污泥,通过富集和划线培养获得两株对AEO有较强降解能力的纯培养A-1和A-2。根据其形态特征、培养特征和生理生化特征等,经初步鉴定为假单胞菌属。 当温度为30℃、pH7.0、接种量为5%、AEO浓度为0.5‰时,培养108小时后,用KI-I2分光光度法测定其降解能力,两株菌株的降解效率均达到90%。 关键词:脂肪醇聚氧乙烯醚;筛选;鉴定;生物降解 Screening and identification of bacterial strains for degrading Alkyl alcohol polyoxyethylene ether Abstract: With the development of modern biotechnology, and more microorganism have a more and more broad range of applications in environment disposal. In this paper, characteristics of bacteria degrading alkyl alcohol polyoxyethylene ether (AEO), which is the most popular used nonionic surfactant in our industry,were studied by screening screening microorganisms which could degrade AEO through slective medium for AEO. Sludge sample were collected from wastewater treatment plant in Qingtan of Changzhou. Two strains named A-1 and A-2 which have the strong competence of degrading were selected from above sludge by enrichment and purification,culture characteristics,and physiological and biochemical reactions, A-1and A-2 was identified as Pseudomonas sp. The degradation ability to AEO of two strains which I had screened was determined by spectrophotometer with KI-I2 solution at the concentration of 1%. The degration rates for AEO both reach to 90% and concentration for AEO at 500mg/L within 108 hours at 30℃, pH7.0,with inoculating quantity at 5%.
