BB-94对大鼠全脑缺血 再灌流损伤后脑水肿作用研究
【摘 要】 目的:观察大鼠全脑缺血-再灌流损伤后脑水肿及BB-94对其的影响。方法:采用Pulsinelli大鼠四血管阻塞模型,缺血15分钟后恢复血流,随机分为正常组、假手术组、再灌流生理盐水组及BB-94治疗组。按规定时间取脑,测定脑组织中脑水含量及EB含量。结果:脑缺血-再灌流损伤时,再灌注3h后脑水含量开始增高,到24、48h达到高峰,BB-94能在24、48h明显降低脑水含量,EB含量在灌注后3、48h处明显增高,BB-94在3、48h处明显降低其含量。结论:脑缺血-再灌流损伤导致脑水肿的发生,人工合成的基质金属蛋白酶抑制剂BB-94能减轻这种脑水肿程度,起脑保护作用,提供了一个新的治疗窗口。

【Abstract】 Objective:To investigate the effect of BB-94 on brain edema after cerebral ischemic reperfusion injury in rats.Methods:The four-vessel occlusion model of Pulsinelli was sed and reperfusion was started at 15min after ischemia. The rat were randomly divided into 4 groups including Normal、Sham-operated、N.S. reperfusion and BB-94 reperfusion. After samples were taken from the brain on time, the water content and EB extravasation were observed.Results:The water content: there was no significant difference between normal group and sham-operated group; it increased gradually after 3 hours of reperfusion and was maximal at 48 hours in N.S. reperfusion. It was significantly lowered in BB-94 group at 24、48 hours. EB extravasation: there was no significant difference between normal group and sham-operated group; it increased at 3、48hours in N.S. reperfusion group. Only after 3 and 48 hours of reperfusion, BB-94 group was apparently lowered than N.S. reperfusion group.Conclusion:When cerebral ischemic reperfusion injury occurred, the water content and EB extravasation of brain tissue were increased. However, BB-94, a synthetic matrix metalloproteinase inhibitor, could decrease the degree of brain edema and extend the therapeutic window.
