摘要检测,该研究为食品安全检测提供了新的研究思路。关键词:黄曲霉毒素B1;量子点荧光微球;免疫层析试纸条;定量检测IIIABSTRACTAflatoxinB1(AFB1)isoneofthemostcompellingmycotoxinsandlistedasaGroupIcarcinogenbytheInternational AgencyforResearchinCancer.Many countrieshaveestablishedregulationstogoverntheAFB1levelinagriculturalproductstoavoidoverexposureofhumansandanimals.Inthepresentstudy,highly luminescentquantumdotbeads(QBs)wereusedforthefirsttimeas immunochromatographicstripsignalprobefordetectionofsmallmoleculeand establishedaquantitativedetectionmodelbasedonFIT/FICfordetectionofAFB1in maize.TheQBs were synthesizedbyencapsulatingCdSe/ZnSquantumdots(QDs),theaveragediameterofthesesubmicrobeadsis247nm±13nmandforsamenumberof QB/QDparticles,luminescenceintensityofQBswaspresumably2863timeshigher thanQDs.Theoptimalparametersofthestripwereasfollows: Thesamplepadsweresaturatedwith20mmol/Lsodiumboratebuffer(pH8.0)containing1.0%(w/v)BSA, 0.25%Tween-20,and0.1%(w/v)NaN3.1mgofQBswereconjugatedto150μg anti-AFB1ascites.TheAFB1-BSA(0.4mg/mL)anddonkeyanti-mouseIgG antibodies(1mg/mL) weredispensedontotheNCmembrane astest andcontrol lines. Inaddition,pHvalue,methanolcontentofthesampleextract,andinterpretationtimecouldinfluencethesensitivityandreproducibilityoftheQB-ICAsensor,immunereactionkineticswasusedtoexploretheinfluenceoftheenvironment.Results indicatedthatthequantitativedetectionmodelbasedonFIT/FICcouldelaboratetheeffectsoftheabovefactors.Theregressionequationcouldberepresentedbyy=-0.28ln(x)+1.25,whereyisthecompetitiveinhibitionrateandxistheAFB1concentration.TheIC50oftheQB-ICAsensorwasachievedat13.87±1.54pg/mL(n=3),whichissignificantlyhigher(39-fold)thanthatoftheQDsasasignalprobe(IC50=0.54±0.06ng/mL).Besides,theQB-basedICAsensorexhibiteddynamiclineardetectionofAFB1inmaizeextractfrom5pg/mLto60pg/mL.ThespecificityoftheQB-ICAsensorwasevaluatedbyrunningfourstructurally relatedanalogs(AFG1,AFG2,AFM1,andAFB2)andotherfivecommonmycotoxins (citrinin,patulin,ochratoxinA,deoxynivalenol,andzearalenone).Theresultsshow thatthedevelopedmethodexhibite
