AbstractTissueengineeringwhichutilizestheprincipleandmethodofengineeringandLifeScienceaimstostudytheformationandregenerationofbodytissues/organsandconstructasubstitutewithbiologicalactivity,whichisembeddedinthebodysoastorepairorimprovepartorthewholeofthehumanbodytissue/organfunction.Thispaperfocusonthedevelopmentofnewmechanicalloadbioreactoranditsapplicationinordertostudytheinfluenceofmechanicalstimulusonthegrowthofthecellsunderthethree-dimensionalscaffoldsinvitro.Withtheaimtomakecellsinvitroproliferation,differentiation,andincreasethemechanicalpropertiesofcell/scaffoldstructures,Itisofgreatsignificancefortheconstructionoftissuesubstitutes,guidingclinicaldiagnosisandtreatment,improvingthelevelofhumanhealthandeconomicdevelopment.Firstly,akindofnewbioreactorwhichcanprovidemechanicalstimulationtocellsinvitroisdevelopedinthispaper.Takingthespecialenvironmentalrequirementsforcellscultureintoconsideration,thispaperdesignsthestructurefromthefollowingfouraspects:thepiezoelectricceramicdynamiccompressionloading,steppermotordynamicshearloading,thedifferentialscrewmechanismoffine-tuningcapabilities,andcanbeusedfortrainingmaterialelasticmodulus.Inordertoverifytherationalityandfeasibilityofdevicestructure,thewholestructure;thedifferentialscrewmechanismandthepurebottompartsbasedonfiniteelementsarerespectivelyanalyzedbasedonthesoftwareANSYSWORKBENCH.Secondly,themechanicstestingmodeldeviceisestablished.Basedonthismodel,theDataCollectingSystemwasdesignedandthedynamicapparentelasticmoduluswastestedandanalyzed.ThestraingaugewasappliedtosimplifytheexperimentalsystemandincontrastoftheDataCollectingSystem,fivekindsofmechanicalloadingmodesforcellsculturearefinallydesigned.Intheaspectofcellculture,thecollagenwasusedtodosurfacemodificationonPLLAscaffolds,thentheanalysisofscaffoldmodifiedresultsweredonethroughcellcompatibilityexperiment.Theosteoblasts,culturedinthevitro3-dementionalscaffolds,wasdonethedynamicmechanicalloadexperimentbytheapplicationsofthemechanicalloadtypebioreactorwhichcanbe
