Effects ofGhrelinontheOvineOocytes DuringinvitroMaturationAbstractGhrelinwasdiscoveredbyJapanesescientistKojimain1999,whichisanew endogenousbrain-gutpeptidemadeof28kindsofaminoacid.Itisanaturalligardof growthhormonesecretagoguereceptor.Combinedwithitsreceptorithascomprehensive physiologicalfunction.SotheresearchprogressofGhrelinisahot pointthat people concernednow.Withdeepresearchofanimalcellprojectandembryoproject,thereportofanimal oocytematurationfactorismainlytheactionprincipleofsomegenetichormone.So,asa naturalligardofgrowthhormonesecretagoguereceptorwehavenotknownwhetherit participatedinthematurationofovineoocyte,whetheritexertedactionthrouthclassical MAPKsignalpassageaboutcellgrowth.Becauseoocytepartlyapoptosisinthecourseof maturation,theexpressionofanti-apoptosisgene-Bcl-2andproapoptosisgene-Bax decidedwhetherfemalereproductivecellwasalive.Nowwehaven’tknownthe relationshipbetweenGhrelinandBcl-2,Baxinthematurationcourseofoocyte.Aimat theseunknownquestions,mypaperesploredtherelationshipbetweenGhrelinandMAPK signal,Bcl-2,BaxinthematurationcourseofovineoocytethroughaddingGhrelinand specialantagonistofGHS-R-D-Lys3-GHRP-6invitromaturationsystemofovineoocye.The results areasfollows:ExpressionofGHS-RintheovineoocyeGHS-R wasmeasuredintheovineoocytethroughRT-PCR.TheinfluenceofGhrelinonthematurationofovineoocyteGhrelinhaspromotionactionontheFirstPolarBody getting–outrateandnucleus mature rateofoocyte,relyingonconsistency.D-Lys3-GHRP-6hasnoeffect ontheFirstPolarBodygetting–outrateand nucleusmature rateofoocyte.ThepromotionactionofGhrelininvitromaturationofoocytecanbeobstructed byD-Lys3-GHRP-6.Themorecompleteits antagonismtoGHS-Ris,themoreobvious the promotionactionofGhrelinwasobstructed.SoitprovedthatGhrelinpromotedoocyte maturationbycombiningwithGHS-R.TheeffectofGhrelinonexpressionofMAPK(ERK1/2),p90rsk mRNAof ovineoocytewas examinedbyreal-timePCR.GhrelinhasascendingactiontotheexpressionofMAPK(ERK1/2),p90rskmRNA ofoocyte,relyingondosageandtimeobviously.D-Lys3-GHRP-6hasn
