英文摘要Different irritantson the ratphagocytecapacity andIntracellular calcium concentration [ Ca2+]iAbstractObjectiv:Theresponseofmacrophagesandneutrophils,playsanimportantrolein theinflammatoryprocess.ThecurrentstudyfoundthatfMLP,PMAandLTB4- inducedperitonealmacrophagesandneutrophilsandthetwocellsinintracellularfree calciumconcentration changes[Ca2+]iphagocyticfunction, such as different styimulus.Intracellularfreecalciumconcentrationandmeasuredthe resting peritoneal macrophages andneutrophils[Ca2+]ilevels andreactivityoffMLP, PMA, LTB4.Method:Abdominallavageandchargeabdominalpermeate,afterwashing, macrophagesandneutrophilsisolatedbydensitygradientcentrifugation.1,Usingof fluorescencemicroscopyandphasecontrastmicroscopyandneutrophilphagocytic cellsoffluorescentmicrospheresandyeastphagocyticcapacitytest.2,Theseparation ofphagocyticcellswithFura-2/AM(finalconcentrationofload1h5μmol/L), washedtoremovedidnotenterthecellswithFura-2/AM,theintracellularion analyzer(dual-wavelengthfluorescencespectrophotometerdollars),thedynamic changesintheexcitationwavelengthof340nmand380nm,emissionwavelengthof510nmof conditions todetermine the cell[Ca2+]i.Results:1,Restingstate,thefluorescentmicrospheresmacrophagephagocytic capacitysignificantlythanneutrophilsisrelativelylarge;fMLP,PMA,LTB4induced enhancementofmacrophageandneutrophilphagocytosisoffluorescentmicrospheres. 2,Restingstate,macrophageswithinthe[Ca2+]ihighconcentrationinneutrophils; differentconcentrationsoffMLP,PMA,LTB4intheroleof[Ca2+]ichangesina concentrationdependentmanner;inducedbyLTB4.[Ca2+]iwasatransientincrease, shortduration,aboutabout1minafterthereturntotherestingstateofconcentration; bythefMLPinduced[Ca2+]iwasalsoatransientincrease,butthelongerthedurationofitsof[Ca2+]iconcentrationslowlydecreasedwithtimeandconcentration dependent;highconcentrationsoffMLPtostimulate,about10minafterrecoveryto therestingstateconcentration;withPMAinductionof[Ca2+]iincreasedthesharpest, the duration ofmost long downward trend observed
