MethodsThe survival and proliferation of HL60 cells were determined by MTT.The cellDNA ploidy distribution and apoptotic rate were measured by flow cytometry (FCM), and DNA ladder.The differentiation induced potence was determined by NBT. The anticancer and apoptosis potence induced by flavonoids detected by TUNEL. ResultsThere was significant inhibition of the cel1 surviva1 and proliferation of cancer cells by the extracts. An apoptosis peak appeared after treated with 3d and apoptotic rate was 38.3%, morphological change and DNA ladder of apoptosis was observed. Kudzu flavonids showed weak potent to induce differentiation. Daidzein and puerarin which regard as Kudzu flavonoids main effect ingredient have weak apoptosis induced potent, but daizein showednbsp; strong differentiation induced potent. ConclusionKudzu flavonoids could significantly inhibit growth of human leukaemia cancer cells HL60 line through inducing apoptosis.   Key words:Kudzu;Flavonoids extracts;Apoptosis;Differentiation   摘要:目的观察葛根黄酮提取物对HL60细胞的影响及作用机制。方法用MTT测定对HL60细胞增殖的影响;用流式细胞仪检测细胞周期变化及凋亡情况;采用NBT比色法检测诱导HL60细胞分化的能力;DNA Ladder检测HL60细胞的凋亡特征及效果;用TUNEL检测体内诱导凋亡能力。结果葛根黄酮提取物有较强抑制HL60细胞生长和增殖能力,处理3d可出现明显的凋亡特征,凋亡率可达38.3%,其诱导分化能力弱,葛根黄酮提取物的主要成分葛根素和大豆甙元的诱导凋亡能力弱,但大豆甙元具有较强的诱导HL60细胞分化的能力,葛根黄酮提取物有体内诱导凋亡抗肿瘤能力。结论葛根黄酮提取物可通过诱导细胞凋亡抗人白血病HL60细胞的生长。   关键词:葛根;黄酮提取物;凋亡;分化   0引言   葛根为常用中药,是豆科植物野葛[Pueraria lobata(Willd.)Ohwi]和粉葛(Pueraria homsonii Bentn)的肥大块根,在我国除新疆、青海及西藏外遍布全国各地。葛根中除含有约占新鲜葛根19%~20%的葛根淀粉外,主要
