proposedanimprovedcellsorterusingtheadjustable-interfacemode,whichhaspotentialtoimprovepurity,throughputandcellviability.Inthecellstimulation,weestablishedachemicalsignalgeneratorbythepinchedmodeforchemicalstimulationofasinglecelllocatedintheanalysischannel.Systemevaluationrevealedthattherisetimeandthefalltimeofthesolutionexchangewere19.8±0.8msand20±1msrespectively.Asaproofofconcept,adenosine-triphosphatestimulationofHeLacellswasdemonstratedbyinvestigatingP2Y-IP3signalingpathwaymediatedcalciumrelease.Furthermore,preliminarystudieshavebeenmadeonthefull-channelmodeandtheadjustable-interfacemodeforcellstimulation.Thefull-channelmodehaspotentialforchemicalstimulationofmultipleindividualcellsintheanalysischannelsimultaneously.Theadjustable-interfacemodeenablescellstimulationinaspatio-temporallycontrollablemannerwithatemporalresolutionoflessthan43msandaspatialresolutionof3.4±0.2μm,whichcanbeusedtoinvestigatesub-cellularcelldynamicsorcellcommunication.Inthedropletpurification,weintegrateddropletgenerationandsortingonasinglechipforafluorescence-activateddropletsorter.Dropletgenerationwasachievedwithathroughputof18droplets/s;dropletsortingwasrealizedbytheadjustable-interfacemodewithbothpurityandrecoveryrateofmorethan99%.Moreover,weproposedhydrodynamicgatingbasedonanaqueous-oilmicrofluidicsystemforselectiveencapsulationmicrobeadsintopicoliter-scaledroplets.Theresultindicatedapurityof92%andarecoveryrateof96%.Atentativeresearchonopen-tubularliquidchromatographicseparationwasalsoconductedbythepinchedmode,andthreekindsoffluorescentdyeswereresolvedusingnativepoly(dimethylsiloxane)asasoildphaseandacetonitrileasamobilephase.Insummary,hydrodynamicgatingwerevalidatedasaversatileanduniversalstrategyformicrofluidicsampleintroduction,anditwasexpectedtobeapowerfulplatformtechnologyforbioanalyticalapplicationsoflab-on-a-chip.Keywords:Lab-on-a-chipMicrofluidicchipHydrodynamicgatingSampleintroductionCellanalysisBioanalysis独创性声明本人声明所呈交的学位论文是我个人在导师指导下进行的研究工作及取得的研究成果。尽我所知，除文中已经标明引用的内容外，本论文不包含任何
