基于SPR技术的单样本检测实验装置-生物医学专业论文
ABSTRACT
SPR biosensor technology is becoming one of the most active research fields in the international academic and industrial community. Because it uses wide modern science and technology such as informatics, material science, mechanism, and microelectronics, SPR biosensor technology is gradually maturing and creates a new and promising market. Thus, it is of great theoretical and practical significance to develop a SPR biosensor device with our own intellectual property rights.
In this thesis, the main work is to set up an experiment platform for body fluid analysis based on SPR biosensor, which utilized new optical, mechanical, electronic technology. The theoretical basis of SPR phenomenon was given at first. Then, an SPR biosensor system was designed and set up for experimental research, which included optic system, sensing system and detection system.
The SPR system in this study is based on the angle-scanning detection mode. Its electric circuit was simplified, and its mechanical system was designed by ourselves for good angle-scanning capacity. The reflective light intensity on the photoelectric cell was changed into electric signal, which was subsequently received by the data acquisition system and sent to the computer. Controlling signal from computer was transferred to the controller to drive the whole system. The sensor chip used for SPR biosensor was also fabricated. Its surface was covered by a self-assembled monolayer for the detection of macromolecular interaction. Finally, some experiments have been done to validate our new SPR design.
