Abstract Abstract Objective:To observe the antidepressant effects of ginseng extract,ginsenoside Rg l and ginsenoside Rb l by using behavial tests in acute stress models of mice,and explore the underlying mechanisms of ginsenoside Rg l and ginsenoside Rb l by using molecular biology and electrophysiology methods. Methods:Various doses of ginseng extract,ginsenoside Rgl or ginsenoside Rbl were intragastrically administrated to mice and their effects on the immobility time in the forced swimming test and the tail suspension test were observed,and their effects on the spontaneous activity in the movement counter and the open—field test were also detected.The effects of ginsenoside Rgl and ginsenoside Rbl on the glutamate receptor-mediated excitatory synaptic transmission and y-amino acid receptor— mediated inhibitory synaptic transmission in the pyramidal neurons from the prefrontal cortex slices of mice were examined by using the whole·-cell patch··clamp recording configuration,and their effects on the expression of related synaptic proteins in prefrontal cortex in mice were also examined. Results:Ginseng extract,ginsenoside Rg l and ginsenoside Rb l decreased the immobility time without affecting the locomotor activity and exploratory behavior in depressive mice.Ginsenoside Rgl and ginsenoside Rbl may promote the release of presynaptic glutamate in prefrontal cortex neurons in mice,and increase the glutamate receptor-mediated excitatory synaptic transmission,while having no significant effect on the expression and function of postsynaptic glutamate receptors;Ginsenoside Rg l and ginsenoside Rb l had no significant effect on),-amino acid receptor-mediated inhibitory synaptic transmission,and the expression of glutamate receptors and associated synaptic proteins. Conclusion:Ginseng extract,ginsenoside Rgl and ginsenoside Rb l showed antidepressant effect.In addition,ginsenoside Rgl showed better effect than ginsenoside Rbl,which due to the significant effect of g
