Abstract：Bioengineering class 134 in Life Science College studied the molecular biology experiment under the theacher Junfeng Pan’s guidence. The whole experiment around the “southern hybridization of digoxin mark”as the center,including experiments with high standards(such as amplication cDNA of RT-PCR and Southern hybridization) and fundamental experiments. Fundamental experiments include the plasmid’s DNA extraction,agarose gel electrophoresis,polymerase chain reaction,plant RNA extraction and electrophoresis,wheat genomes DNA extraction,enzyme digestion,electrophoresis analysis,preparation and transferma-
tion of cells. Most experiments went well,but the quality of extracted DNA and RNA was not very good.Additionally,the result of electrophoresis analysis of RT-PCR’s product just showed primer,not target gene.After analysis,we think the reason of failing may be material,it was freezed too long.

2.实验：
2.1 实验材料
暗培养7天的小麦幼苗（黄化苗），含质粒的大肠杆菌P38
2.2 试剂
2.2.1质粒DNA提取
质粒小提试剂盒（天根公司，中国产）
LB液体培养基：胰蛋白胨10g，酵母提取物5g，NaCl 10g，溶解于1000mL蒸馏水中，用NaOH调pH至7.0。高压灭菌20分钟。
LB平板培养基：在每1000mL LB液体培养基中中加入15g琼脂，高压灭菌20分钟。
溶液Ⅰ：50mmol/L葡萄糖，10mmol/LEDTA，25mmol/L Tris-Hcl(pH8.0),冰箱保存，RNA酶要提前加入溶液Ⅰ中；
溶液Ⅱ：0.2mol/L NaOH,1%SDS（碱性的NaOH使蛋白质变性，SDS结合变性的蛋白质）；
溶液Ⅲ：Ph4.8的醋酸钾溶液（5mol/L乙酸钾60ml，冰乙酸11.5ml,水28.5ml）；
TE缓冲液（pH8.0）：10mmol/L Tris-HCl，1mmol/L EDTAl；
无水乙醇和70%乙醇。
2.2.2 质粒DNA酶切及琼脂糖电泳分析鉴定
EcoRⅠ酶（Thermo,美国） λDNA
TBE缓冲液（5×）：用时需稀释10倍
点样缓冲液Loading buffer（10×）：0.25%溴酚蓝，40%甘油
溴乙啶(EB)：10mg/ml溴乙啶 （注意：该试剂具致癌作用，用时要小心） 琼脂糖1%
2.2.3 质粒载体和外源DNA的连接反应
目标DNA片段（P38质粒的PCR扩增产物）
载体（pGEM-T Easy）
2×ligation 缓冲液
T4 DNA连接酶
2.2.4感受态细胞的制备及转化
0.1mol/L CaCl2取20μl PCR产物进行1.2%琼脂糖电泳分析。
