PAGE PAGE 5 Papain-induced Knee Osteoarthritis in Mice with Different Concentrations ABSTRACT Objective: The objective of the research was to compare the knee osteoarthritis (OA) models in mice with different concentrations of papain mixed solution and provide data for exploring pathogenesis, treatments and Chinese herbal medicine research of this disease. Methods: ①eighty Kunming mice , average weight 48g, were randomly divided into four groups of 20 each and given injections into the right knee on the 1st, 3rd, 5th day respetively. The intra-articular injections were composed of 2%, 4%, 8% (w/v) papain and 0.3mol/L L-cysteine at the dose of 0.25ml/kg (experimental groups). The 0.9%Nacl (w/v) with same dose were injected intra-articularly into the right knees of mice in the control group; ②The mice were sacrificed at one week respectively after the initiation of papain injection and this OA models were evaluated through recording the width of knee joints, performing the morphological observation (toluidine bluc staining HE staining) and histological evaluation of articular cartilage. Results: ① swelling, flaxion dysfunction, drapping steps and activities diminishment could be observed in experimental groups, meanwhile with swelling knee joints. ② The degenerative changes were demonstrated in knee joints of mice in all experimental groups, such as thinner articular cartilage, fibrillation and destroyed cartilage matrix, and inflammation, proliferation. All these changes were much worse with increased concentration. ③ Mankin score: There was significant difference between the control group and the three experimental groups (P0.05). There were significantly difference among the three experimental groups in Mankin score(P0.05) and the scores increased with the increasing of concentration. Conclusion(s): Different severityof OA are established through intra-articular injections of 2%, 4%, 8% papain and 0.03mol/L L-cysteine at the dose of 0.25ml/kg. These models are
