实时荧光定量 PCR 检测转基因玉米 Bt176 测量不确定度分析

摘 要:应用实时定量 PCR 方法测定样品中转基因玉米 Bt176 的含量,并从多个方面来分析测量的不确定度。
结果显示,A 类不确定度为 0.0003,B 类不确定度为 0.0007,合成不确定度为 0.001,有效自由度veff 为 272 ,包含
概率 P 为 95%,扩展不确定度 U=0.002 ,测量结果为(0.8±0.2)% 。不确定度评估结果表明,测量不确定度的主要
来源是微量液体转移量的偏差。

Uncertainty in detecting event-specific fragments of genetically modified
maize BT176 by quantitative real time PCR

Abstract: The real-time fluorescent quantitative PCR method was used to detect the event-specific fragments
in a sample of genetically modified maize BT176 and the uncertainty of measurement derived from several as-
pects was analyzed. The results showed that uncertainty of type A and type B was 0.0003 and 0.0007, respectively;
the combined standard uncertainty was 0.001; the effective degree of freedom was 272; the coverage probability
was 95%; the expanded uncertainty was 0.001, and the measured result was 0.8%±0.2%. This result indicated that
the main source of uncertainty was the trace amount of liquid transfer, which should be paid more attention to in
future detection.
