China Oct-2015 摘要 为研究LPS对大鼠乳腺形态结构影响，选择体重相近的泌乳大鼠10只，适应环境后对 其灌注LPS,观察临床症状并采集样本，制作病理组织学切片。结果发现，灌注LPS泌乳 大鼠乳腺组织周围有明显充血，间质增多，腺泡结构相对完整，部分乳腺上皮细胞坏死脱落, 腺泡腔内乳汁减少、淡染呈絮状，且散在少量坏死脱落的上皮细胞，腺泡间散在少量红细胞。 结果表明，LPS能够引起泌乳大鼠乳腺的炎症反应。同时，为培养大鼠乳腺上皮细胞，采取 正常泌乳人鼠乳腺组织，用机械剪切结合消化酶的方法，在体外进行原代乳腺上皮细胞的分 离培养。结果培养出呈典型鹅卵石状的乳腺上皮细胞。本试验为进一步研究LPS致大鼠急 性乳腺炎发病机制奠定了基础。 关键词：LPS大鼠乳腺病理变化乳腺上皮细胞 Abstract In order to investigate the effects of LPS on the morphology and structure of rat mammary gl and, 10 female rats with similar weight were selected, after adapting to the environment, the LPS was infected. After that, the clinical symptoms were observed, the samples were collected, and the pathological slices were made .It was found that LPS instillation of lactation rat mammary tissue a round was congested, interstitial increased, acinar structure is relatively complete, partial breast epi thclial cell necrosis, acinar cavity hypogalactia, pale stained flocculent, and scattered in a small am ount of necrosis of epithelial cells, acinar scattered in a small amount of red blood cells? At the sa me time, in order to cultured rat mammary epithelial cells, use the method of mechanical shear and digestive enzyme with the normal lactating rat mammary gland tissue, to isolate and culture prima ry mammary epithelial cells. The results showed that the cells of mammary epithelial cells were ty pical cobblestone? This experiment lays the foundation for further research on the pathogenesis of acute mastitis induced by LPS in rats.. Keywords: LPS ; rat
