SCOPE

Thisgeneralchapterisintendedtoproviderecommendationsontheminimumqualityattributesforstartingmaterialsusedinthemanufactureofsynthetictherapeuticpeptides.Becauseitisnotpossibletocoverthewidevarietyofavailablestartingmaterialsthatmaybeused,includingaminoacids,protectedaminoacidderivatives,andpeptidefragments,thediscussioninthischapterwillbelimitedtothemostcommonlyusedprotectedaminoacidderivatives(AAD),i.e.,9-fluorenylmethyloxycarbonyl(Fmoc)aminoacidderivatives(includingresin-boundAAD).However,itisintendedthatthegeneralconceptsandguidanceprovidedforthisclassofstartingmaterialsshouldbeappliedtoallpeptidestartingmaterials,whereapplicable.ThischapteristobeusedinconjunctionwithQualityAttributesofSyntheticPeptideDrugSubstances〈1503〉.

INTRODUCTION

Theincreasedinterestinpeptidesasdrugcandidatesduringthepastfewdecadescanbeattributedtoanumberoffactors,includingadvancesinproteomicsandhigh-throughputscreeningtechniques,whichhaveacceleratedthedevelopmentofnewpotentialtherapeuticcandidates.Furthermore,thedevelopmentofefficientsyntheticchemicalstrategies,combinedwithrobustpurificationprocessesandmorepowerfulanddiscriminatinganalyticalmethodologies,havefacilitatedthemanufactureofpeptideswithhigh-qualitystandards.The
