Introduction:

U251gliomacelllineiswidelyusedinthestudyofgliomabecauseofitsreliabilityandreproducibility.However,themolecularmechanismsinvolvedintheprogressionandtreatmentofgliomaarestillpoorlyunderstood.RNAmodificationsplayavitalroleinregulatinggeneexpressionandmodulatingvariousbiologicalprocesses,includingcellgrowth,differentiation,andapoptosis.Mettl9isamemberoftheN6-methyltransferasefamilythatcanincreasethestabilityandtranslationefficiencyofmRNA.However,theroleofMettl9ingliomacellproliferationandapoptosisisstillunclear.Therefore,theaimofthisstudyistoinvestigatetheimpactofMettl9geneontheproliferationandapoptosisofU251gliomacells.

Methods:

1.CellCultureandTreatment:TheU251gliomacellswereculturedinDulbecco’sModifiedEagleMedium(DMEM)supplementedwith10%fetalbovineserum(FBS)and1%penicillin-streptomycinsolution.ThecellswerethentransfectedwithMettl9siRNAorscrambledsiRNAusingLipofectamine2000accordingtothemanufacturersinstructions.

2.CellViabilityAssay:TheU251cellswereseededin96-wellplatesatadensityof5×10?cellsperwell.TheCellCountingKit-8(CCK-8)assaywasusedtoevaluatecellviabilityat24,48,and72haftertransfection.

3.ColonyFormationAssay:TheU251cellswereseededin6-wellplatesatadensityof1×103cellsperwell.Thenumberofcoloniesformedafter14daysofincubationwascountedunderamicroscope.

4.FlowCytometryAnalysis:TheapoptosisrateofU251cellswasassessedbyflowcytometryusingAnnexinV-fluoresceinisothiocyanate/propidiumiodidestaining.
